Human pluripotent stem cells (hPSCs) are attractive sources for modeling human kidney diseases in vitro and kidney regenerative medicine, yet the protocol for differentiation into kidney lineage cells has eluded many investigators over the years. Recently, we have established a highly efficient and specific protocol for generation of SIX2+ nephron progenitor cells (NPCs) and kidney organoids consisting of podocytes, proximal tubules, loops of Henle, and distal tubules, from hPSCs. Our lab utilizes this state-of-the-art technology for studies of human kidney development and injury, genetic kidney diseases, and kidney regeneration.
